Abstract
Bu çalışmada zirai mücadele uygulamalarında sıklıkla kullanılan ve canlılar üzerinde çeşitli toksik etkilere neden olan klorprifos ile güçlü bir antioksidan olarak bilinen curcuminin kalpte oluşturduğu histolojik değişiklikler ışık mikroskobu düzeyinde araştırılmıştır. Bu amaçla kontrol, klorprifos, curcumin ve klorprifos+curcumin olmak üzere 4 grup hayvan kullanılmıştır. Ratlara (Wistar Albino) curcumin ve klorprifos gavaj yoluyla verildikten sonra 15. ve 30. günlerde kardiyak perfüzyon işleminin ardından kalpler çıkarılmıştır. Kalplerin fiksasyonu %10 tamponlanmış nötral formalin ile yapıldıktan sonra rutin histolojik işlemler uygulanmıştır. Preperatlar hematoksilen eozin, periodik asit-Shiff (PAS) ve Masson Trikrom teknikleri ile boyanmıştır. Çalışmadan elde edilen sonuçlara göre; klorprifosun etkisiyle, kalpte periferik çekirdek, sitoplazmik vakuol, hücre infiltrasyonu, stromal fibrozis ve koagülasyon nekrozu gözlenmiştir. Yalnızca curcumin verilen gruplarda hafif sitoplazmik vakuol ve koagülasyon nekrozu dışında kalpte belirgin bir değişiklik olmamıştır. Klorprifos ve curcuminin birlikte verildiği gruplarda ise, sadece klorprifos verilen gruplara göre daha az histolojik değişiklik gözlenmiştir. Bu bulgular, bir serbest radikal süpürücü olan curcumin, klorprifosun serbest radikal oluşumunu indükleyerek kalpte meydana getirdiği oksidatif hasarı azalttığını göstermektedir.Ratların kemik iliklerinden micronukleus tespiti için kolsişin kullanıldı. Çalışmadan elde edilen sonuçlara göre klorprifos uygulanan gruplarda poliploidi, kromozom kırıkları, fragment oluşumu gözlenmiştir. Yalnızca curcumin uygulanan gruplarda kromatit kırığı ve poliploidi dışında genotoksik etki görülmemiştir. Klorprifos ve curcuminin birlikte verildiği gruplarda genotoksik etkilerin azaldığı gözlenmiştir. In this study, histologic changes induced by chlorpyrifos which is the often used in agriculture studies and caused varius toxic effects in life and curcumin which is known as a strong antioxidant, were investigated under light microscopy. For this purpose, four groups of animals were used as curcumin, chlorpyrifos, curcumin+chlorpyrifos and control groups. Curcumin and chlorpyrifos were given via gavage rats (Wistar albino), their hearts were removed at 15 and 30 days after cardiac perfusion. After isolated hearts were fixed 10% neutral formalin, were applied and sections were stained by hematoxilen-eosin (H-E), Masson's trichrome and periodic asit- Schiff (PAS). According to results obtained in our study, histologic changes such as peripheral nucleus, cytoplasmic vacuole, cell infiltration and coagulation necrosis induced by chlorpyrifos were determined. It was seen that using only curcumin didn't cause any obvious differentiation on heart expect cytoplasmic vacuole and coagulation necrosis. When chlorpyrifos and curcumin were given together, the degree of histological changes induced by chlorpyrifos and curcumin, was less than chlorpyrifos alone. These findings show that curcumin, as a free radical sweeper, decreases the oxidative damage that chlorpyrifos causes in heart by induced free radical formation. Colchicine was used for to determination rat's the bone marrow micronucleus. According to results obtained in our study, polyploidi , chromosome fracture and fragments induced by chlorpyrifos were determined. It was seen that using only curcumin didn't cause any genotoxic effect polyploidy and chromosome fracture. When chlorpyrifos and curcumin were given together, genotoxic effect induced by chlorpyrifos and curcumin, was less than chlorpyrifos alone.
