Abstract
Ill ÖZET ASPİRİN VE ANTİOKSİDANT BUTYLATED HYDROXYANİSOLE'ÜN FARELERDE (Mus musculus) KARACİ?ER RNA MİKTARINA ETKİLERİ Bu çalışmada aspirin ve antioksidant butylated hydroxyanisole'un fare karaciğerindeki total RNA miktarına etkileri karşılaştırmalı olarak çalışılmıştır. Aspirin ve butylated hydroxyanisole farelere intraperitonal enjeksiyonla verildikten sonra 2., 4., 8., 12. ve 24. saatlerde, servikal dislokasyonla öldürülerek karaciğerleri çıkarıldı. Daha sonra bu karaciğerlerden homogenizasyon ve differansiyal santrifügasyon teknikleri kullanılarak RNA'lar izole edildi. İzole edilen RNA'ların miktarları orsinol yöntemi ile belirlendi. Çalışmalarda; Aspirin etkisi ile total karaciğer RNA düzeyinin yükseldiği, butylated hydroxyanisole etkisi ile belirgin bir artış olmamasına karşın aspirin + butylated hydroxyanisole etkisi ile RNA düzeyinin arttığı belirlenmiştir.
IV ABSTRACT THE EFFECTS OF ASPIRIN AND ANTIOXSIDANT BUTYLATED HYDROXYANISOLE ON THE LEVEL OF LIVER RNA IN MICE {Mus musculus). In this study, the effects of aspirin and antioxsidant butylated hydroxyanisole were studied comparatively on the total amount of RNA in the mice liver. Aspirin and butylated hydroxyonisole were given to mice by intraperitoneal injection (i.p.) and after the injection at the 2'd, 4th, 8th, 12th and 24th hours mice were killed by servical discolation; and their livers were removed, and then total RNA were isolated from these livers by using the homogenization and differantial centrifugation techiniques. Amounts of isolated RNA s were determined by orcinol assay. As a result, it was determined that total liver RNA levels effectively increased by the effect of aspirin and aspirin + butylated hydroxyanisole, however the levels of RNA increased very little by the effect of butylated hydroxyanisole individually.
