Abstract
Streptomyces violaceusniger KÜMESİ (Clade) ÜYELERİNİN RİZOSFERDEN İZOLASYONU VE KARAKTERİZASYONU ÖZET Bu çalışmada, ağaç ve çalı formundaki Acasia dealbata, Albizia distachya, Colutea arborescens, Gladitsia triancanthos, Medicago arborea, Sophora japonica, Spartium junceum, Tipuana tipu ve Wisteria sinensis baklagillerin rizosferinden, kök patojeni olan funguslara karşı antagonistik etkisi olduğu bilinen Streptomyces violaceusniger kümesi üyelerinin izolasyonu ve nümerik taksonomisi yapılmıştır. İzolasyonda dilüsyon plak tekniği uygulanmış olup cyloheximide (50 ug/ml), nystatin (50 ug/ml) ve rifampicin (0.5 ug/ml) ilaveli nişasta-kazein agar üzerine 0.2 mi toprak dilusyonu ilave edilen izolasyon plakları 25°C'de 14 gün inkübe edildi. Saf kültürleri yapılan ve oatmeal agar üzerine transfer edilen toplam 305 izolat, oatmeal agar üzerindeki pigmentasyon özelliklerine göre 22 büyük (5-49 suş), 12 küçük (2-4 suş) ve 9 tek üyeli kümeye ayrılmıştır. Streptomyces violaceusniger kümesi üyelerinin karakteristik pigmentasyon özelliklerini gösteren 35. ve 37. renk grupları üyeleri, melanin pigmenti üretip üretmediklerini belirlemek amacıyla pepton yeast-extract iron agar üzerine transfer edilmiş, bunlardan melanin pigmenti üretmeyen ve spor zincir morfolojisi spiral olan 75 izolat test organizması olarak belirlenmiştir. Nümerik analizler için 75 test izolatı, Streptomyces violaceusniger kümesi üyesi olan 7 tip Örneği ve 6 duplike sustan oluşan toplam 88 test susuna, besinsel, biyokimyasal, degredasyon ve fizyolojik testlerden oluşan toplam 94 test uygulanmıştır. Elde edilen test sonuçlan, X-Taxon programında Ssm-UPGMA analizleri ile değerlendirilmiştir. Son veri matriksine göre NTSys-pc programıyla yapılan analiz sonucu, % 85 benzerlik değeriyle 6 büyük (5-14 suş), 4 küçük (2-4 suş) ve 21 tek üyeli küme oluşmuştur. İİ ISOLATION AND CHARACTERISATION OF MEMBERS OF THE Streptomyces violaceusniger CLADE FROM RHIZOSPHERE ABSTRACT In this study, we have isolated the members of the clade of Streptomyces violaceusniger, which is known to have antagonistic effect on root infecting fungi, from the rhizosphere of legume of Acasia dealbata, Albizia distachya, Colutea arborescens, Gladitsia triancanthos, Medicago arborea, Sophora japonica, Spartium junceum, Tipuana tipu ve Wisteria sinensis in the form tree or shrub, and made a numerical taxonomy of them. We have used dilutions plate technique during the process of isolation, and isolation plates, which were added 0.2 ml soil dilution on starch-casein agar supplemented cycloheximide (50 ug/ml), nystatin (50 ug/ml) and rifampicin (0.5 Ug/ml), were incubated at 25°C for 14 days. 304 isolates, which were subcultured and transferred on oatmeal agar, were divided into 22 major (5-49 strains), 12 minor (2-4 strains) and 9 single membered clusters according to their pigmentation features on oatmeal agar. The members of colour groups 33rd and 37th which have characteristic pigmentation features of the members of Streptomyces violaceusniger were transferred on pepton-yeast extract iron agar in order to determine whether they produce melanin pigments, and those which did not produce melanin pigments and had spiral spore chain morphology determined 75 isolates as test organisms. For numerical analysis, we have applied total 94 tests consisting of nutritional, biochemical, degredation and physiological tests on 88 test strains including 75 test isolates, 7 type strains which is a member of Streptomyces violaceusniger clade and 6 duplicated organisms. The results obtained were evaluated on X-Taxon programme with Ssm-UPGMA analysis. According to the last data matrix, the result of the analysis which was done using NTSys-pc program, 6 major (5-14 strains), 4 minor (2-4 strains) and 21 single membered clusters were formed which were % 85 with a high similarity values.
