Abstract
Bu çalışmada, öncelikle bütünüyle katı-hal kompozit amonyum seçici sensörhazırlanarak durgun ortamda test edildi. Sensörün potansiyometrik performansınıniteleyen özellikler bilgisayar kontrollü ölçüm sisteminde test edildi. Sensörün ana iyonyanında girişim yapan iyonlara karşı seçicilik katsayıları ?ayrı çözelti metodu? ilehesaplanarak tespit edildi. Hazırlanan sensör 1x10-1-5x10-5 mol.L-1 konsantrasyonaralığında ana iyon çözeltisine karşı doğrusal davranış gösterdi. Sensörün cevapzamanının kısa (<8s) ve kullanım ömrünün uzun olduğu belirlendi (3 ay).L-amino asit oksidaz enziminin, tek adımda enzim tutturma metodu iletamamıyla katı-hal kompozit NH4+-seçici sensör yüzeyine bağlanmasıyla, amino asitbiyosensörü hazırlandı. Hazırlanan amino asit biyosensörünün performansı farklı pH vedeğişik konsantrasyonlardaki tampon çözeltileri kullanılarak test edildi. Geliştirilenamino asit biyosensörü, uzun kullanım ömrü, düşük maliyette üretim, minyatürizeedilebilme ve kısa cevap zamanı gibi önemli üstünlüklere sahiptir. Geliştirilenbiyosensör; hareketli ortamlarda kullanılabilen mikrolitre ölü hacme sahip dedektörhücresi üretmeye dolayısıyla kromatografik sistemlerde amino asit dedektör olarakkullanılmaya elverişlidir. Optimum şartlar altında amino asit biyosensörü, 1x10-1 __M konsantrasyon aralığında bir çok amino aside karşı doğrusal cevap sergilerken,1x10bazı amino asitlere karşı doğrusal cevap sergilememektedir.Anahtar Kelimeler: Amonyum Seçici Sensörler; Ayrı Çözelti Metodu; TamamıylaKatı-Hal Kompozit Elektrot; Amino Asit Biyosensör.

The potentiometric performancecharacteristics of the prepared sensor was tested by using a computer-controlledpotentiometric system. Selectivity coefficients of the sensor againts interfering ions incomparison with main ion was calculated by ?separate solution method?. The preparedsensor exhibited a linear response between concentrations 1x10-1 and 5x10-5 mol.L-1solutions of main ion. The sensors exhibited short response time (<8s), long life time (3months).Amino acid biosensor prepared by immobilisation of L-amino acid oxidaseenzyme onto all solid-state composite NH4+-selective sensor with single step enzymeimmobilisation method. The characteristics of the prepared amino acid biosensor wereexamined in several buffer solutions at different concentrations and pH values.Improving biosensor have important characteristics like long life-time, easily built at alow cost, micro-construction and short response time. The technology used here for theconstruction of the sensor allows production of detector cell with microliter deadvolume using under flowing conditions however may use as aminoacid detector in thechromatography systems.
