Publication: Karadeniz Bölgesinde Yetişen Çeşitli Bitkilerde Bazı Anyonların Kapiler Elektroforez ile Tayini
Abstract
Kapiler elektroforez ve yöntem optimizasyonu için bir Box-Behnken kemometrik tasarım ile seçilen bazı bitki ve bitki çaylarında nitrat (NO-3), nitrit (NO2-) ve bromat (BrO3-) iyonlarının eşzamanlı olarak belirlenmesi için yeni bir yöntem geliştirilmiştir. Çalışmada yürütücü tampon olarak pH'sı 3,5 olan 60 mM fosfat çözeltisi, optimum çalışma koşulları için: 40 °C sıcaklık, -22,5 kV uygulama gerilimi, 50 µm çapta 61,5 cm uzunlukta (etkin kapiler uzunluğu 53,5 cm) kapiler kolon ve 50 mbar' lık basınçta 30 s enjeksiyon süresi uygun görülmüştür. Nitrat, bromat ve nitrit için tayini sınırları 0,49 mg L-1, 1,25 mg L-1 ve 0,11 mg L-1 iken doğrusallık değerleri sırasıyla 0,998, 0,996 ve 0,999 olarak belirlenmiştir. Anyonların analizi ise 5 dakikalık sürede gerçekleştirildi. Geliştirilen yöntem çeşitli kuru bitki yapraklarına,çeşitli kuru çay yapraklarına, laboratuvardan alınan musluk suyuna, arıtılmış musluk suyuna ve marketlerden alınan doğal kaynak sularına uygulanmıştır.
A novel approach to the simultaneous determination of nitrate (NO3-), nitrite (NO2-) and bromate (BrO3-) ions in some selected plant and tea samples using capillary electrophoresis and a Box-Behnken chemometric design for method optimization was developed. Optimal conditions of 60 mM phosphate buffer solution at 3.5 pH, capillary temperature of 40 C, -22.5 kV applied voltage, total capillary length of 61.5 cm (effective capillary length 53. 5 cm) with 50 µm i.d, and 50 mbar injection pressure for 30 s were selected. The limits of detection for nitrate, bromate and nitrite were 0.49 mg L-1, 1.25 mg L-1 and 0.11 mg L-1 while linearity values were determined as 0.998, 0.996 and 0.999 respectively. The detection of the anions was achievable within 5 min of analysis. The method was then applied to the analysis of different real samples namely dry plant leaves, dry tea leaves, tap water from the lab, purified tap water and two different brands of bottled mineral water sold in markets in Samsun city, Turkey
