Abstract
Bu çalışmada farklı Aktinobakteri üyelerinin polifazik taksonomisi gerçekleştirilip olası yeni Aktinobakterilerin literatüre kazandırılması amaçlanmıştır. Sınıflandırılma çalışmalarının hız kazanması, farklı habitatlardan Aktinomiset çalışılmasının önemini arttırmıştır. Türkiye'den Ürgüp-Göreme yöresinden temin edilen, olası Aktinobakteri suşlarının toprak izolasyonu 8 farklı seçici besiyeri kullanılarak gerçekleştirilmiştir. 530 Aktinobakteri suşu izole edilmiş ve bu izolatların 157'sinin 16S rRNA gen bölgesi dizi analizi yapılmıştır. Baz dizi analizleri Chromas Pro 1.7.6 yazılımı kullanılarak gerçekleştirilmiş ve filogenetik ağaçlar, neighbour-joining, maximum likelihood ve maximum parsimony algoritmaları ile MEGA 7.0 programında oluşturulmuştur. Muhtemel yeni tür olduğu belirlenen suşların verilerinin Genbank'a (NCBI=National Center for Biotechnology Information, US) depoziti, nümerik analizleri, atpD, gyrB, recA, rpoB ve trpB birleştirilmiş dizileme analizleri, detaylı yağ asidi ve menakinon analizleri, tüm hücre analizleri ve spor morfolojisi görüntülemeleri elde edilmiştir. Amycolatopsis sp. AG28 (DSM 104280) izolatının tüm polifazik tanımlama çalışmaları tamamlanarak Amycolatopsis cappadoca DSM 104280T olarak ülkemiz topraklarından yeni tür olarak literatüre kazandırılmıştır. 16S rRNA gen bölgesi dizi analizlerine göre yeni tür oldukları belirlenen diğer Aktinobakteri üyelerinin de polifazik çalışmaları ivedilikle tamamlanarak ülkemizden bakteriyal sistematik literatürüne kazandırılacaklardır.
In this study, polyphasic taxonomy of different members of actinomycetes will be performed and possible novel species are aimed to gain literature. Acceleration of classification studies has increased the importance of studying actinobacteria from different regions. Putative Actinomycetes strains were isolated from soil samples collected from different ecological habitats belonging to Turkey, Ürgüp-Göreme, using 8 different selective media. 530 actinobacteria strains were isolated and 157 of these isolates were performed to 16S rRNA gene sequence analysis. Obtained sequence data were aligned in Chromas Pro 1.7.6 programme and phylogenetic relationships of isolates were analysed in MEGA 7.0 and dendrograms were constructed by using neighbour-joining, maximum likelihood and maximum parsimony algorithms. Deposit of the sequence data of the strains to Genbank identified as likely to be new species, numerical analysis, atpD, gyrB, recA, rpoB ve trpB multilocus sequence analyses, whole cell assays, analyses of fatty acids and menaquinons, spore morphology analysis was made. All polyphasic identification studies of Amycolatopsis sp. AG28 (DSM 104280) isolate were completed and Amycolatopsis cappadocia DSM 104280T as a new species from the territory of our country has been brought into the literature. According to 16S rRNA gene sequence analysis of identified novel species of other Actinobacteria members will immediatly be gained to bacterial systematic literature from our country soil samples by completing polyphasic studies.
